Abstract
In this study, polyethylene glycol (PEG) based microgels were fabricated and evaluated their potential as a drug carrier. Polyethylene glycol sebacic acid diacrylate (PEGSDA) microgels were synthesized using emulsion polymerization and Texas red dextran (TRD) or bone morphogenetic protein-2 (BMP-2) was incorporated into the PEGSDA microgels. The drugs were homogeneously distributed throughout the PEGSDA microgels. TRD and BMP-2 release profiles from the microgels showed much less initial burst release compared to the PEGSDA bulk gels, thus it is possible to achieve sustained release of drugs of interest using the microgels. The bioactivities of BMP-2 released from the microgels were measured by proliferation and alkaline phosphatase (ALP) activity of W-20-17 (bone marrow stromal cell line) cells cultured in the presence of the released biomolecules. As a results, there were any detrimental effects on proliferation of the cells, and ALP activity of bone marrow stromal cells treated with BMP-2 released from PEGSDA microgels showed well maintained bioactivity of BMP-2. In addition, the bone marrow stromal cells were cultured on the surface of the microgels and the cells were well attached to the microgels, which shows another possible use of the microgels as a cell delivery system.

본 연구에서는 생분해성 고분자인 polyethylene glycol(PEG) sebacic acid diacrylate(PEGSDA) 기반 마이크로젤을 합성하고 이를 이용하여 약물전달시스템으로서의 응용가능성에 대하여 살펴보았다. 먼저 에멀젼 중합을 통하여 PEGSDA 마이크로젤을 합성하고 모델약물인 Texas red dextran(TRD)와 bone morphogenetic protein-2(BMP-2)를 이용하여 마이크로젤 내에 함침하고 그 분포를 측정한 결과 젤 내에 균일하게 분포되는 것을 확인하였다. 약물방출속도를 측정한 결과 기존의 PEGSDA 벌크젤에 비하여 초기방출속도를 추가적으로 감소시켜 서방형 용출이 가능한 것을 확인하였다. 또한 용출된 BMP-2의 생활성을 측정하기 위하여 W-20-17 세포를 이용하여 마이크로젤로부터 용출된 BMP-2의 세포증식 및 alkaline phosphatase(ALP) 활성을 측정하였다. 그 결과 PEGSDA 마이크로젤은 세포독성이 발견되지 않았으며 용출된 BMP-2의 경우, 그 활성을 잘 유지하는 것을 관찰하였다. 마지막으로 마이크로젤 표면에 세포를 배양한 결과 세포가 잘 부착되는 것을 확인함으로써 세포전달시스템으로서의 가능성도 타진하였다.
