Abstract

Thermo-responsive hydrogels consisted of methylcellulose (MC) and xanthan gum (XT) could be used as an injectable reparing material to regenerate bone defect. In this study, the hydrogel containing xenogenic bone graft materials (BGM) was investigated for enhancement of bone defect regeneration in vivo. MC-XT aqueous solution containing BGM exhibited sol-to-gel transition at physiological temperature. In a cranial defect animal model studies, the hydrogel containing BGM improved the bone regeneration significantly in comparision with the untreated group and the hydrogel only group. As shown in the result of micro-CT and immunohistochemical staining, newly formed bone tissues were clearly observed at 4 weeks post-treatment in the hydrogel containing BGM group. The results indicate that the thermoresponsive MC-XT hydrogel containing BGM was an useful injectable material for bone defect regeneration in vivo.


골 재생을 유도하기 위해 주사가 가능한 메틸셀룰로스(methylcellulose, MC)와 잔탄검(xanthan gum, XT)으로 구성된 온도감응형 하이드로젤을 제조하였다. 본 연구에서는 생체 내에서 골 결손 재생을 향상시키기 위해 골 이식재(BGM)가 포함된 하이드로젤을 연구하였다. BGM이 함유된 MC-XT 수용액은 생리학적 온도에서 졸-젤 전이를 보였다. 두개골 결손 동물 모델 연구에서, BGM을 함유한 하이드로젤은 무처치 그룹 및 하이드로젤만 처리한 그룹과 비교하였을 때 뼈 재생을 상당히 개선시켰다. Micro-CT 및 조직학적 결과에서 볼 수 있듯이, BGM을 포함한 하이드로젤에서는 치료 4주 후에 새로 형성된 골조직이 분명하게 관찰되었다. 결론적으로 BGM을 함유한 온도감응형 MC-XT 하이드로젤은 생체 내에 골 결손 재생을 위한 유용한 주사 물질임을 확인하였다.
